What is SS-31?
SS-31 (Elamipretide) is a synthetic tetrapeptide with the sequence D-Arg–Dmt–Lys–Phe–NH₂ (where Dmt stands for 2′,6′-dimethyltyrosine), representing a model compound from the Szeto-Schiller peptide group. This compound was developed and characterized in the early 21st century as part of research into peptides capable of selective localization within mitochondrial membranes. Due to its resistance to enzymatic degradation and good solubility in an aqueous environment, SS-31 has become a useful model for studying peptide signaling in the context of mitochondrial function.
In experimental studies, SS-31 has been used to analyze mechanisms related to mitochondrial homeostasis, interactions with cardiolipin, and the cellular response to oxidative stress. Its effect on the modulation of processes occurring in the inner mitochondrial membrane—including elements associated with the respiratory chain—as well as on the reorganization of mitochondrial structures in in vitro and in vivo models has been described. Attention has been drawn to its interactions with membrane lipids and its indirect influence on redox processes under experimental conditions.
Due to its precisely defined chemical structure and high reproducibility of observed biological effects in preclinical models, SS-31 is used as a research tool in molecular biology, cell biology, and experimental pharmacology. In scientific literature, it is also utilized as a reference peptide for evaluating mitochondrial processes and for validating analytical methods used in the study of short bioactive peptides.
